ODRŽIVOST ANTIMIKROBNE AKTIVNOSTI MIKROKAPSULIRANOG ETERIČNOG ULJA KADULJE

ABSTRACT:
Essential
oil of south Dalmatian sage (Salvia offcinalis L.) was microencapsulated
in gelatin-accacia microcapsules in 1986. Antimicrobial activity and retention
time effect on Bacillus subtilis was tested in 1987. and the inhibition
zones obtained were 17 mm. The microcapsule samples were stored at room temperature
in glass bottles for 15 years. Antimicrobial activity of microcapsules was examined
on B. subtilis after storage period and similar results were obtained.
In ten measurements the inhibition zones were in range between 16-17 mm, with
an average of 16.5 mm. Kinetics of essential oil release from a microcapsule
(diameter 50 wm) was examined by microantibiogram on microculture with 8. subtilis
(37 °C) and observed under the light microscope during 36 hours. The
observed microcapsule swells after a very short time (2 min), while after 1
hour droplets of essential oil (1.5-2_m) emerge at the microcapsule surface.
Droplets of essential oil are seen around the microcapsule surface (1 mm) after
6-8 hours, and after additional eventually after 8 hours there is a clear inhibition
zone of 1.2 mm around the microcapsule. Results show that microencapsulation
of sage essential oil is very suitable for preservation, in context of stability
and unchanged antimicrobial activity during long period of time.

SAŽETAK:
Eterično ulje
kadulje (Salvia officinalis L.), podrijetlom iz južne Dalmacije, mikrokapsulirano
je 1986 godine u mikrokapsule od želatine i arapske gume. Antimikrobna aktivnost
i retencijsko vrijeme djelovanja na Bacillus subtilis objavljeno je 1987
godine, kada je izmjerena zona inhibicije 17 mm. Mikrokapsule su pohranjene
u staklene bočice i držane na sobnoj temperaturi 15 godina. Nakon tog vremena
ponovno je ispitana antimikrobna aktivnost na B. subtilis, koja je ostala
nepromjenjena. U 10 mjerenja izmjerene su zone inhibicije 16-17 mm, odnosno
prosjecna zona inhibicije je iznosila 16.5 mm. Kinetika otpustanja eteričnog
ulja iz mikrokapsule (dijametra 50 _m) ispitana je utvrđivanjem mikroantibiograma
na mikrokulturi s B. subtilis (37 °C) i praćena je pod mikroskopom
36 sati. Bubrenje mikrokapsule zapaža se vec nakon 2 minute, a kroz 1 sat kapljice
eteričnog ulja (1.5-2 _m) izbijaju na površinu mikrokapsule. Nakon 6-8 sati
(retencijsko vrijeme) kapljice eteričnog ulja nalaze se raspršene u okolici
mikrokapsule na udaljenosti od 1 mm, a nakon 8 sati u okolisu mikrokapsule vide
se čiste zone inhibicije 1.2 mm. Rezultati pokazuju da mikrokapsuliranje eteričnog
ulja kadulje može odrzati stabilnost i aktivnost 15 godina na sobnoj temperature.
